New pilot proficiency test: NGS-based analysis of circulating tumor DNA (NGSctDNA)
With the newly initiated pilot interlaboratory test for the NGS-based investigation of circulating tumor DNA (ctDNA), the RfB is expanding its range of services in the field of molecular oncology. The aim of the proficiency test is to evaluate the quality and standardization of analytical methods in the context of liquid biopsy under practical conditions.
The focus is on the investigation of therapy-relevant tumor genes using next-generation sequencing (NGS) from circulating tumor DNA. Participating laboratories have the opportunity to review and further develop their methodological expertise and bioinformatics evaluation within the framework of an external quality assurance process.

Key data on the NGSctDNA proficiency test
- Registration: 11.08.2025 to 29.08.2025
- Trial period: 29.09.2025 to 01.11.2025
- Sample dispatch: Week 39
- Participation fee: free of charge (only shipping costs according to the shipping price list)
- Number of participants: max. 15 laboratories (first-come-first-serve, extension reserved)
- Transmission of results: via the participant account on www.rfb.bio
Content and requirements
Various samples will be made available as part of the proficiency test. The assessment is based on laboratory-specific sequencing data and documented process steps. Among other things, information is requested on:
- Sample processing (e.g. QC, library production, sequencing)
- Data analysis (bioinformatics evaluation and filter criteria)
The following file formats are required for evaluation:
- BED file of the sequencing panel used
- FASTQ files
- Sample sheet or Excel file with index mapping
- BAM Files
- VCF files (related to hg38) incl. indication of the filter criteria used
A standardized BED file of the therapy-relevant tumor genes covered in the trial can be provided if required.

Participation is only possible via a registered online account at www.rfb.bio . As a pilot project, the proficiency test is subject to special conditions – there is no legal entitlement to participation. The terms and conditions of the Foundation for Pathobiochemistry and Molecular Diagnostics apply.
The proficiency test is aimed at diagnostic laboratories with experience in the field of liquid biopsy and NGS analysis. Early registration is recommended.
